I want my science museum to have a small room that's only accessible by going through an unmarked door at the back of another exhibit.
Inside there will be a sign to the effect of "Congratulations! You found the secret room!" but I'm not sure what else should be in the room...

Go digital with it, make it a backwards-projected touchscreen.
That way you can filter what drawings and wrtiting stays on it, and if someone draws a dick or something you can erase it without ruining the rest of the wall.

See, I was thinking that if we're going technological, it might be better to have a computer in there with a camera on it. You can take your picture (and add your name if you like), then view pictures of everyone else who's found the secret room and left their pictures there.
It would be cheaper than touchscreen walls (remember, only a fraction of museum goers should be seeing this anyway, so it's not worth going all-out) and it would be a bit more visual.
...I'm wondering if this is even really feasible. It's a building open to the general public; once someone spreads word of the secret room it won't be a bloody secret anymore.

I was thinking of putting it in the Perception exhibit.
The Perception exhibit has stuff that's basically meant to mess with your mind--there's an Ames room inside, and the entrance to the exhibit from the main hallway is built with slanted walls and ceiling in such a way that you feel like you're leaning over when in fact you're walking perfectly straight and level.
I figure if we stick a seemingly innocuous door somewhere like that, where everyone's gonna be preoccupied with something more interesting, only the truly observant will find it.

So with a marker we'd end up with a room full of dicks, and with a computer we'd end up with a computer that's turned on 24/7 despite being used very rarely.

A Durasig would be cool, but I was hoping for something a bit older. And given the limited amount of space, and I should probably go for one with 8" lenses; I don't think another 12-incher would really fit.
Though there's always ped signals...eventually I'd like to get my Eagle Mark IV mounted on some kind of stand and then attach a ped signal to the same stand...

AFIAK Siemens still produces signals with the Durasig wordmark on the back, but doesn't market them under that name. Basically they're just using the old molds and not giving a fuck about any old trademarks on them.
