Been binging on Nostalgia Critic lately and that's where I heard it. (I also heard it when I watched the movie the first time, but that was way back when it was first released, so I'd forgotten about it.) Here are some alternate lines which may also have worked.
Man is a most complex simple creature: see what he weaves, and how base his reasons for doing so.
Deacon is the "motherfucker." By attempting to be the Blood God and making himself invincible, he is trying to "iceskate uphill," an endeavor that while seemingly rewarding, is treacherous. If Deacon fails he'll fall down the hill. If he gets up there, he'll still fall, because Deacon wears iceskates (the EDTA in the serum explodes vampire blood, and becoming the Blood God won't take that away. Blade's observation is basically a dramatic warning against hubris, except given in a profane, devil-may-care way.
I got at least part of that when I heard it, like that it's a way of saying some people just insist on trying to do the impossible, but it still sounds kind of odd, at least to me. It doesn't ruin the movie or anything, it's just odd.
