I expect my Let's Plays to be video walkthroughs of the game, discussing how the game works and is structured. Basic LP should be a video walkthrough of all the features of the game acquirable in one playthrough. More specific LP types show of specific features, such as an LP dedicated to sequence breaking, or glitching, or 100%ing a game.
The only exception is a blind LP, which is about the player's reactions to things.
My favorite LPs are the ones that are part-walkthrough and part-fanfic. It differentiates it from the standard walkthrough, gives it a character of its own.
This is a good example. It's an LP of Princess Maker 2, an RPG-ish game where you have to train a girl to become a princess of sorts. The LP mixes it up by making Gendo the girl's father and adding in some crazy dialogue.
This is probably my all-time favorite. It's an LP of Alice in Wonderland on the Commodore 64. The split between walkthrough and fanfic is pretty even, and Alice and the Author are written adorably.

But I watch LPs rarely enough that it doesn't matter anyway.
Basically, you have a knowledgeable player, who provides a video walkthrough with voice commentary, mostly tips on how to get through the game and descriptions of what's going on from a player's experience perspective.
This is because, when I watch an LP, generally speaking I want to find out about the game (which I likely have never played before), and I want the commenter to introduce me to the game and its gameplay features. Your mileage may vary if your purpose of watching an LP is different.

raocow has a genuine talent for making his commentary go thud in a very entertaining way. That's also not even close to all he does, he does spend a good chunk of his time talking about the game too.
